Abstract: Laboratory equipment for sampling is often expensive and sometimes cannot be used for animals. We needed to collect samples of saliva and urine from even-toed ungulates, concretely from Bactrian camels (Camelus bactrianus) kept in zoological gardens and domestic cattle (Holstein dairy cows), for the research focused on reproduction which was realised between April 2010 and February 2012. So we tried to find cheap supplies for sampling. The sampling of urine by plastic cup mounted to rod is quite common for urine sampling. We used clear plastic PET cups with volume of 0.5 litre mounted by wire to broom handle for urine collection from females camels. When the female started to urinate, we approached toward her and collected the urine to the cup. We collected by this way 78 samples of urine from 13 female camels. Most of animals had no problem with the collection. Only three females (16.6 %) were “shy” and stopped urination when someone got closer to her. But it was usually possible to collect urine from these females as well. The sampling of small amount of saliva was realised by plastic coffee stirrers. Saliva production was stimulated by dry bread in case of camels or by the hand inside the mouth in case of cattle. We collected successfully 623 saliva samples from 11 camels and 408 saliva samples from 8 cows. We also collected saliva of camels in bigger amount for hormonal assays. We used the bigger plastic spoon for sampling. The handle of the spoon was plunged to the mouth when a camel was trying to attempt the bread and saliva got stuck in the depression of the spoon handle. Saliva was removed by coffee stirrer to test tube and the process was repeated several times until we collected more than 2 ml of foamy saliva. We collected successfully 422 samples from 16 camels. Camels behaved to be fed from hand had no problem with the collection. We concluded that plastic catering supplies are adequate and cheap equipment for sampling. Abstract: The accurate heat and pregnancy detection is one of the decisive factors of successful livestock breeding. Available methods in management of livestock reproduction are time-consuming and expensive. The saliva sampling is simple and noninvasive procedure, which is applicable in untamed animals and the presence of veterinary is not needed. The phenomenon of crystallization of body fluid is known over 60 years. The saliva crystallization was mainly described in women. Only two studies focused on saliva crystallization in animals are available. The aim of this thesis was confirmation of presence of saliva arborization in cattle and observation of changes in crystallization during synchronized cycle and pregnancy. The Giemsa stain was additionally tested as tool for better searching of crystals. The eight cows of Holstein breed were included into research and 408 samples were microscopically evaluated in total. The samples were obtained daily from the first application of preparation of synchronization till 34thday after insemination. The crystals were scored according to following system: 0 = none, D = dotted, BL = branch-like, FIL = fir-like, FEL = fern-like, BL+FIL = mixed branch-like and fir-like, BL+FEL = mixed branch-like and fern-like, FIL+FEL = mixed fir-like and fern-like, BL+FIL+FEL = mixed branch-like, fir-like and fern-like, A = atypical. All mentioned types of crystallization were confirmed in tested animals as well as the changes during whole observed period. At the time of artificial insemination were presented only two types of arborization (BL+FIL and BL+FIL+FEL). The predominant types of crystallization, noticeable after insemination (FIL+FEL, FIL, BL), were marked only in pregnant animals. It was confirmed, that sample quality influenced the density of crystals. Some of crystalline patterns, detected in cattle saliva, were similar with crystals described in other studies focused on crystallization of body fluids..

The pregnancy diagnosis in half-tamed animals is difficult. The aim of this research was to verify the Cuboni reaction and barium chloride test in Bactrian camels (Camelus bactrianus) and their usage as a non-invasive tool for diagnosis of pregnancy from the urine. The urine of nine adult female camels from Prague, Plzen and Bratislava zoological gardens was collected to the half-litre plastic cups fasted to 1.5 meters long rod in one or two months intervals. Forty one samples were evaluated during September 2010 to February 2011. The samples of urine were stored in a refrigerator and the analyses were carried out within 14 days after sampling. The Cuboni reaction was performed by the State Veterinary Institute Prague. The minimal amount of the sample was 20 ml and the reaction was evaluated like positive, negative, and dubious. The barium chloride test was done in a laboratory of the university by 1% solution of barium chloride. One, five, ten, and fifteen drops of solution were added to the 5 ml of urine and the reactions (presence of white precipitate) were described. Both tests are based on chemical reaction of oestrogen in urine. Three females were pregnant and give a birth later. The Cuboni test of urine in these females showed in 86.7% positive and in 13.3% dubious reactions. The positive reaction was noted even 215 days before parturition. We did not find any false negative reaction but we noted positive and dubious results in non-pregnant females as well. There is high probability that camel urine reacts positive even if the female is in the heat due to higher oestrogen levels than in mares of domestic horses. We did not find any clear relationship between the reproductive status of females and the results of barium chloride tests. There was also no correlation between results of Cuboni and barium chloride reactions. The Cuboni test can be a useful tool for pregnancy diagnosis in camels during the second half of gestation. If the urine capture is repeated 10-14 days after first sampling, the false positive result, due to the possibility of the heat, can be eliminated. The research of Cuboni tests will continue and will be more focussed on the first half of gestation, and the time when the first positive reaction of urine will occur. ČERNÍNOVÁ, R. – HABEROVÁ, T. – KOLÁČKOVÁ, K. Podtyp: Příspěvek ve sborníku (mimo kategorie RIV); Agonistic behaviour of African ungulates kept in mixed species exhibits. 2011. Breeding of African ungulates in common exhibits is very popular for its advantages from the perspective of visitors as well as zoos. Despite this fact, the formation of mixed group can be problematic. The aim of this study was the evaluation of agonistic behaviour, especially determination of the most frequent initiator and recipient species of aggressive interactions and of the most common winner species. We also focused on the most common interactions, situations in which the aggressive behaviour appeared, and other factors which could influence this behaviour such as the presence of calves in herds. The observation was realised in six zoological gardens in the Czech Republic - Brno, Dvůr Králové, Ostrava, Plzeň, Praha and Zlín (in total 123 animals). The herds of these zoos were of similar compositions with mixed herds of giraffes, zebras and antelopes. Ostriches were also included because they are often bred together with African ungulates. The agonistic behaviour was recorded ad libitum and data were tested by nonparametric tests (Kruskal-Wallis ANOVA, Mann-Whitney test) and contingency tables. It was recorded 153 intraspecific and 354 interspecific interactions during 180 hours of observation. We found significant differences among the frequency in aggressive encounters in particular species (H [10, 210]=55.47, p<0.01). Ostriches (Struthio camelus), gnus (Connochaetes taurinus), and Scimitar horned oryxes (Oryx dammah) attacked other species significantly more often than blesboks (Damaliscus dorcas phillipsi), and lechwes (Kobus leche). The frequency of interspecific conflicts was higher than intraspecific interactions in most species. The most affected species was especially zebra but also other species. The less affected species were giraffes and gnus. The types of aggressive behaviour during actions and reactions were significantly species-specific. The most frequent actions were chasing (48.9 %) (mainly in giraffes, ostriches, zebras and gnus) and horns attack (29.8 %) (especially in oryxes, elands and gnus). The most common reactions were passive as running (61.3 %) and walking away (23.1 %). Elands and gnus won all conflicts that they initiated but also Scimitar horned oryxes, giraffes, beisas, and zebras succeeded in more than in 90.0 % of initiated interactions. Gnus, elands, and giraffes also won the interactions as affected species in 50.0, 41.7, and 28.6 % of cases. Other affected species lost the majority of conflicts. The attack frequency was significantly higher in herds without calves and most interactions occurred during grazing and feeding. In general, the most problematic species were Scimitar oryxes, gnus, and ostriches. It is not possible to form a mixed-species herd without problems but species such as lechwe, blesbok, nyala, giraffe or eland can be recommended for breeding in common enclosures..

ČERNÍNOVÁ, R. – HABEROVÁ, T. – KOLÁČKOVÁ, K. Podtyp: Příspěvek ve sborníku (mimo kategorie RIV); Prostorové rozdělení afrických kopytníků chovaných ve smíšených výbězích v lidské péči. 2011. Chov zvířat ve společných výbězích je dnes hojně rozšířen, především se jedná o chov kopytníků. Na základě jejich prostorového chování (rozdělení) můžeme určit, zda jednotlivá zvířata ve výběhu tvoří opravdu smíšená stáda nebo se jednotlivé druhy drží odděleně od ostatních druhů. Cílem této práce bylo vyhodnocení prostorového rozdělení afrických kopytníků chovaných ve společných výbězích v lidské péči. Předpokládaly jsme, že jednotlivé druhy kopytníků se budou držet spíše odděleně, čili nebudou vytvářet smíšená stáda. Faktory, které mohou prostorové rozdělení zvířat ve stádě ovlivnit jsou především počet krmných míst, dale pak velikost výběhu a možnost pastvy ve výběhu. Sledování kopytníků probíhalo v roce 2010 na šesti stádech vybraných zoologických zahrad v České republice (Zoo Praha, Zoo Dvůr Králové, Zoo Plzeň, Zoo Brno, Zoo Ostrava and Zoo Zlín). Pro tuto studii byla vybrána stáda s minimálním množstvím tří afrických druhů a nejméně dvou jedinců každého druhu (celkem 12 druhů, 121 zvířat). Stáda byla podobného složení, přičemž se kladl důraz na stáda složená z žiraf, zeber a antilop. Pro prostorové chování kopytníků jsme použily metodu scan sampling. Pro každé stádo byla vytvořena tabulka zaznamenávající vzdálenosti mezi jednotlivými zvířaty v intervalu 10 minut (celkem pozorováno 180 hodin). Vzdálenosti, které se počítaly v zebřích délkách, byly rozděleny do tří kategorií (do 2 zebřích délek, do 5 a nad 5 zebřích délek). K vyhodnocení jednotlivých vzdáleností byl použit neparametrický test Kruskal-Wallisova ANOVA, který signifikantně prokázal, že se jednotlivé druhy ve výběhu drží odděleně a netvoří smíšená stáda, tedy do vzdálenosti dvou a pěti zebřích délek se zdržovala zvířata především stejného druhu, čímž se potvrdila stanovená hypotéza..
